A multipeptide melanoma vaccine combination consisting of twelve MHC class I-restricted melanoma peptides (12MP) and six MHC class II-restricted melanoma helper peptides (6MHP). The 12MP component is designed to stimulate CD8+ cytotoxic T lymphocyte (CTL) responses against shared melanoma antigens, including gp100, MART-1, Tyrosinase, MAGE-A1, MAGE-A3, and NY-ESO-1. The 6MHP component provides cognate CD4+ T cell help, which is hypothesized to enhance the development and durability of circulatory memory CD8+ T cells. Clinical data from the Mel44 trial (NCT00118274) demonstrated that patients receiving the 12MP + 6MHP combination had improved long-term clinical outcomes and better preservation of memory CD8+ T cells compared to those receiving 12MP with a non-cognate tetanus helper peptide.
