131I-huA33 + capecitabine is a combination therapy consisting of a humanized monoclonal antibody (huA33) labeled with radioactive iodine (iodine-131, or 131I) and the oral chemotherapeutic agent capecitabine. The huA33 antibody specifically targets the A33 antigen, which is highly expressed in over 95% of human colon cancers and in normal intestinal epithelium but not other tissues. When labeled with 131I, huA33 delivers targeted radiotherapy directly to colorectal cancer cells. Capecitabine is an oral prodrug that is metabolized to fluorouracil (5-FU), a cytotoxic antimetabolite that inhibits DNA synthesis in rapidly dividing cells. The combination aims to provide synergistic effects by delivering both targeted radiation and chemotherapy directly to metastatic colorectal cancer lesions while minimizing toxicity to normal tissues. This approach has been evaluated primarily for safety, tolerability, pharmacokinetics, biodistribution, immunogenicity, and preliminary efficacy in patients with metastatic colorectal cancer[1][2][5][7].
