Overview

17,18-Epoxyeicosatetraenoic acid (17,18-EpETE or 17,18-EEQ) is an endogenous lipid mediator and cytochrome P450 (CYP) epoxygenase metabolite of the omega-3 fatty acid eicosapentaenoic acid (EPA). It acts as a potent anti-inflammatory and anti-allergic agent, primarily targeting G protein-coupled receptor 40 (GPR40, also known as free fatty acid receptor 1 or FFAR1) to suppress neutrophil infiltration, reduce pro-inflammatory cytokine production (such as IL-6 and IL-8), and inhibit mucin secretion. Additionally, it has been shown to activate sphingosine-1-phosphate receptor 1 (S1P1) and open large-conductance calcium-activated potassium (BK) channels. 17,18-EpETE has been investigated in preclinical research for its potential to treat airway inflammation (e.g., asthma, chronic rhinosinusitis), contact hypersensitivity, and to mitigate local inflammatory side effects associated with mRNA-LNP vaccines. While 17,18-EpETE itself is primarily used as a research tool due to rapid metabolism by soluble epoxide hydrolase, stable synthetic analogs like OMT-28 have been developed for clinical evaluation in cardiovascular and mitochondrial diseases.
