NAN, or 17-Cyclopropylmethyl-3,14β-dihydroxy-4,5α-epoxy-6α-(indole-7-carboxamido)morphinan, is a novel small molecule opioid receptor modulator being investigated for the treatment of opioid use disorder. It functions primarily as a μ opioid receptor (MOR) antagonist, preventing agonist-induced MOR internalization and intracellular Ca2+ release. Additionally, NAN acts as a low efficacy partial agonist for G-protein activation at both MOR and δ opioid receptors (DOR), and exhibits relatively high efficacy agonism at κ opioid receptors (KOR). Preclinical studies suggest it may produce fewer withdrawal signs compared to naltrexone and does not induce fentanyl-like discriminative stimulus effects.
