Overview

**19(T2)28z1xx TRAC T cells** are autologous chimeric antigen receptor (CAR)-T cells engineered to target CD19-expressing B-cell malignancies. These cells are created by integrating a CD19-targeted CAR construct into the T-cell receptor alpha constant (TRAC) locus of patient-derived T-cells. The construct features a modified signaling domain with only one functional immunoreceptor tyrosine-based activation motif (ITAM), termed "1XX," which is designed to calibrate activation potential and reduce toxicity compared to traditional constructs with three ITAMs. This modification aims to enhance efficacy, persistence, and safety by reducing the risk of severe cytokine release syndrome and neurotoxicity while maintaining potent anti-tumor activity. The therapy is under investigation primarily for relapsed or refractory diffuse large B-cell lymphoma (DLBCL), with early clinical trials showing promising response rates and durability of remission[4][5][6].
