**1C3 + 1C11** is a cocktail of two human monoclonal antibodies derived from Ebola survivors, designed as a pan-ebolavirus therapeutic. **1C3** binds asymmetrically to the trimer apex (GP head region) of the Ebola virus glycoprotein (GP), contacting residues 115, 117–120, 124, and 172 across three protomers with unique 1:3 stoichiometry, while **1C11** targets the fusion loop at the GP base, bridging adjacent protomers and interacting with the N-linked glycan at position 563. The combination exhibits synergistic neutralization (combination index 0.63 against EBOV pseudovirus), fully neutralizes escape mutants of either antibody alone, and provides 100% protection in mouse models of EBOV/BDBV-GP infection and complete protection in nonhuman primates against lethal EBOV and SUDV challenges at low doses (e.g., 25 mg/kg each at days 4 and 7 post-exposure).[1][3][7][10][11]
