Overview

2'-3' cGAMP (2',3'-cyclic GMP-AMP) is an endogenous cyclic dinucleotide (CDN) produced in mammalian cells by the enzyme cyclic GMP-AMP synthase (cGAS) in response to cytosolic double-stranded DNA. It acts as a potent second messenger that binds to and activates the Stimulator of Interferon Genes (STING) receptor with high affinity. This activation triggers the TBK1/IRF3 and NF-κB signaling pathways, resulting in the induction of type I interferons (IFN-β) and various pro-inflammatory cytokines. While primarily utilized as a research reagent, 2'-3' cGAMP is being extensively studied for its potential as a vaccine adjuvant and as an immunotherapeutic agent in oncology and antiviral therapy. It is structurally distinct from the bacterial canonical 3'3'-cGAMP and is currently available from multiple chemical suppliers for laboratory use.
