Overview

2'-Deoxyuridine (dU) is a nucleoside that commonly occurs as a DNA lesion, arising from cytosine deamination and the incorporation of dUMP by DNA polymerases, processes frequently observed in cancer. The repair of these dU lesions in genomic DNA is primarily mediated by the base excision repair pathway, initiated by Uracil-DNA Glycosylase 2 (UNG2). Modulating pathways involving 2'-deoxyuridine, such as through UNG inhibitors, shows potential for influencing inflammatory responses in tumors. Additionally, 2'-deoxyuridine is identified as a cerebrospinal fluid metabolite, with its levels causally linked to intracranial aneurysms. Derivatives of 2'-deoxyuridine are also being investigated as promising antimicrobial agents, demonstrating inhibitory activity against pathogens like Mycobacterium tuberculosis and Gram-positive bacteria.
