Overview

2'3'-cGAM(PS)2 is a **synthetic bisphosphorothioate analog** of the endogenous cyclic dinucleotide (CDN) 2'3'-cGAMP, designed to be more stable and potent than the natural ligand. It acts primarily as a **STING (stimulator of interferon genes) pathway agonist**, inducing **type I interferon production** and thereby influencing innate immune responses[1][2][5]. In addition to canonical STING-mediated effects, recent studies indicate that 2'3'-cGAM(PS)2—and 2'3'-cGAMP generally—can robustly promote cell migration in certain cancer cells through a **STING-independent pathway** involving direct activation of the small GTPase Rab18, leading to downstream induction of the transcription factor FosB[3][4]. The bisphosphorothioate modification prevents rapid enzymatic degradation, enhancing the molecule’s stability for research and potential therapeutic applications[2]. Main uses are in immunotherapy research and mechanistic studies involving innate immunity and cell migration.
