Drug intelligence / Profile preview
2,4-Diamino-6-hydroxypyrimidine (DAHP) is a selective and specific small molecule inhibitor of GTP cyclohydrolase I (GTPCH1), the rate-limiting enzyme in the de novo biosynthesis of tetrahydrobiopterin (BH4). By depleting systemic BH4, DAHP promotes the uncoupling of constitutive nitric oxide synthases (NOS), leading to decreased nitric oxide production and increased generation of reactive oxygen species such as superoxide. In preclinical research, DAHP is widely utilized as a pharmacological tool to induce endothelial dysfunction, pulmonary hypertension, and diastolic dysfunction mimicking heart failure with preserved ejection fraction (HFpEF) in animal models.
