Overview

**2,4-Dinitrophenol** is an organic compound (C6H4N2O5) and member of the dinitrophenol family, formerly used industrially in wood preservatives, herbicides, dyes, sulfur dye manufacture, and explosives (including as a TNT precursor). Clinically, it was prescribed as one of the earliest weight-loss drugs in the 1930s due to its ability to raise basal metabolic rate through mitochondrial uncoupling, but was banned for human use due to risk of fatal hyperthermia, multi-organ toxicity, and deaths. As a protonophore, 2,4-dinitrophenol disrupts ATP synthesis in mitochondria by collapsing the proton gradient, leading to energy dissipation as heat. It is not approved for pharmaceutical use, is classified as hazardous, and continues to be used illegally for weight loss purposes[1][3][5].
