It acts as an insulin sensitizer by serving as an agonist of peroxisome proliferator-activated receptor gamma (PPARγ), a nuclear receptor that regulates gene expression involved in glucose and lipid metabolism. This mechanism enhances insulin sensitivity and improves glucose uptake in peripheral tissues. While 2,4-thiazolidinedione itself is primarily used in research settings rather than directly as a pharmaceutical agent, it serves as the core scaffold for clinically approved drugs such as pioglitazone and rosiglitazone for type 2 diabetes mellitus.
