Overview

**Description:** 2',3'-Dehydro-2',3'-Deoxy-Thymidine 5'-Diphosphate (D4T-DP) is a synthetic pyrimidine nucleotide analog and the diphosphate metabolite of stavudine (d4T), a nucleoside reverse transcriptase inhibitor. It features a diphosphate group linked to the ribose moiety of thymidine that lacks hydroxyl groups at positions 2 and 3. As an experimental compound and not an approved drug itself for clinical use in humans or animals as of June 2025[1][4], it is primarily studied as an intermediate or active metabolite in antiviral research targeting HIV reverse transcriptase. The compound has been used in structural studies to understand activation by nucleotide diphosphate kinase and inhibition of HIV reverse transcriptase—including drug-resistant variants—suggesting its relevance for antiretroviral drug development[9].
