2'-deoxyadenosine 5'-triphosphate (dATP) is a naturally occurring nucleotide analog of adenosine triphosphate (ATP). While primarily known as a substrate for DNA synthesis, dATP has been identified as a potent cardiac inotrope. It acts as a superior substrate for cardiac myosin compared to ATP, increasing the rate of cross-bridge formation and power output in cardiac muscle. Specifically, dATP destabilizes the super-relaxed (SRX) state of myosin heads, recruiting them into an active, force-generating state. This mechanism is being investigated as a therapeutic strategy for heart failure, particularly right ventricular (RV) dysfunction associated with pulmonary hypertension, where a high proportion of myosin is trapped in the non-force-producing SRX state.
