Overview

2,4-diamino-5(3',4'-dichlorophenyl)-6-methylpyrimidine (DDMP) is a diaminopyrimidine folate antagonist. Initial clinical experience with DDMP involved its administration to tumor patients, often in combination with folinic acid. The drug demonstrated dose-limiting myelosuppression, particularly thrombopenia, and skin rashes. Objective responses were observed in patients with epidermoid cancer of the head and neck, epidermoid cancer of the lung, and melanoma. It was typically administered orally.
