2-(2-chlorophenylamino)-5-(4-fluorophenyl)-1,3,4-thiadiazole (also known as compound B3) is a synthetic small molecule and nonsteroidal aromatase inhibitor. It was designed as a potential therapeutic agent for hormone-dependent breast cancer by targeting the estrogen biosynthesis pathway. The compound features a 1,3,4-thiadiazole core substituted with a 2-chlorophenylamino group and a 4-fluorophenyl group. In preclinical evaluations, it demonstrated selective cytotoxic activity against estrogen-dependent MCF-7 breast cancer cells (IC50 ≈ 52-55 µM) while showing minimal activity against estrogen-independent MDA-MB-231 cells. Molecular docking and dynamics simulations suggest that the compound binds to the active site of the aromatase enzyme, forming interactions characteristic of established inhibitors, such as mimicking the binding mode of native inhibitors and interacting with the heme group.
