Overview

2-Aminoquinazolin-4(3H)-one is a small molecule belonging to the class of quinazolinamines. It has been identified as a promising lead compound in several therapeutic areas. Notably, it exhibits potent antiviral activity against SARS-CoV-2 and MERS-CoV with low cytotoxicity and has also shown efficacy as an inhibitor of malaria digestive vacuole plasmepsins I, II, and IV. In cell-based assays, derivatives have demonstrated growth inhibition of Plasmodium falciparum with submicromolar potency. The mechanism of action involves inhibition of viral entry for coronaviruses and inhibition of plasmepsin enzymes for antimalarial activity. Derivatives are under investigation for improved pharmacokinetics and oral bioavailability[3][5].
