Overview

3,19-4-bromobenzylidene-isoandrographolide is a synthetic small-molecule acetal derivative of isoandrographolide, which is a diterpene lactone structurally related to andrographolide (the major bioactive constituent of Andrographis paniculata). Developed by researchers at Tuskegee University, Sri Sathya Sai Institute of Higher Learning, and the National Institute of Pharmaceutical Education and Research (NIPER) Hyderabad, the compound is being investigated as a potential therapeutic for breast cancer and leukemia. In preclinical studies, it demonstrated potent in vitro anticancer activity against MCF-7 and BT-549 breast cancer cells, as well as MOLT-4 leukemia cells. Its mechanism of action involves the induction of apoptosis, cell cycle arrest at the G1-phase, and the loss of mitochondrial membrane potential, though a specific single molecular biological target has not been identified.
