Compound 5c is a novel, disubstituted flavone glycoside synthesized via click chemistry (copper-catalyzed azide-alkyne cycloaddition) for the potential treatment of glioblastoma (GBM). A derivative of 3,7-dihydroxyflavone, this small molecule is characterized by its high molecular weight and significant lipophilicity, which presents challenges for standard administration. To overcome solubility issues and facilitate delivery across the blood-brain barrier, it is encapsulated in PEG-PLA polymersomes functionalized with a T7 transferrin-mimetic peptide (HRPYIAH). In preclinical models, the compound has demonstrated the ability to significantly reduce the metabolic activity of U-251 human glioblastoma cells while maintaining cytocompatibility with normal fibroblast cell lines. The research was described in a 2025 publication, and the compound is currently in the preclinical stage of development.
