3-{[4-(but-2-yn-1-yloxy)phenyl]sulfonyl}propane-1-thiol is a synthetic small molecule belonging to the class of benzenesulfonyl compounds and sulfones. It features a butyn-yloxy group attached to a phenyl ring with a sulfonyl linkage and a terminal thiol group on the propane chain. This compound was developed as part of research into potent inhibitors of tumor necrosis factor-alpha converting enzyme (TACE), also known as ADAM17, which is involved in the cleavage of membrane-bound TNF-alpha and other substrates. The compound (notably referred to as "compound 8b" in literature) demonstrated excellent in vitro potency against TACE/ADAM17 with good selectivity over matrix metalloproteinases (MMPs), including MMP2, MMP7, MMP8, MMP9, and MMP13. Its mechanism involves binding to the catalytic site of TACE via its thiol moiety chelating the active site zinc ion. There are no approved clinical indications or marketed products containing this molecule; it remains an experimental tool compound for biochemical studies on metalloprotease inhibition[6].
