3BNC117-LS + 10-1074-LS is a combination of two long-acting, broadly neutralizing antibodies (bNAbs) designed for the treatment and prevention of HIV-1 infection. **3BNC117-LS** is a human monoclonal antibody that targets the CD4 binding site of the HIV-1 envelope glycoprotein gp120, while **10-1074-LS** targets the V3 glycan supersite on the same protein. Both antibodies incorporate the 'LS' mutation (M428L/N434S) in the Fc region, which increases their affinity for the neonatal Fc receptor (FcRn). This modification enhances the recycling of the antibodies, significantly extending their half-life and allowing for less frequent dosing compared to their parent molecules. The combination is primarily being investigated for its ability to maintain viral suppression and target persistent viral reservoirs in individuals living with HIV who are currently on antiretroviral therapy (ART).
