Drug intelligence / Profile preview
3H-1,2-dithiole-3-thione (D3T) is a sulfur-containing dithiolethione and a potent antioxidant originally found in cruciferous vegetables. It acts primarily as an activator of the nuclear factor erythroid 2-related factor 2 (Nrf2) transcription pathway, which induces the expression of phase II cytoprotective and antioxidant enzymes, such as glutathione S-transferase and NADH quinone oxidoreductase 1 (NQO1). D3T has demonstrated significant anti-inflammatory, neuroprotective, and chemopreventive properties in preclinical models. It has been investigated for its therapeutic potential in various autoimmune and inflammatory conditions, including experimental autoimmune encephalomyelitis (EAE), psoriasis, lupus nephritis, and ischemic stroke, where it suppresses pathogenic Th1 and Th17 cell differentiation and inhibits NLRP3 inflammasome activation.
