Overview

4'-Bromoresveratrol (4'-BR) is an experimental small molecule and a synthetic brominated derivative of the natural polyphenol resveratrol. Unlike its parent compound, which acts as a SIRT1 activator, 4'-bromoresveratrol is a potent dual inhibitor of the sirtuin deacetylase enzymes SIRT1 and SIRT3. It is primarily investigated for its anti-cancer potential, particularly in melanoma and gastric cancer, where SIRT1 and SIRT3 are frequently overexpressed. Preclinical studies in human melanoma cell lines and patient-derived xenograft (PDX) models have shown that 4'-BR inhibits cell proliferation, induces apoptosis, and reduces metastatic potential. It is currently in the preclinical stage of development and is widely used as a research tool.
