4'-demethylepipodophyllotoxin is a semi-synthetic derivative of podophyllotoxin and a critical intermediate in the production of the anticancer agents etoposide and teniposide. Developed by XinKe Pharmaceutical, this small molecule functions as a DNA topoisomerase II inhibitor. By stabilizing the transient complex between the enzyme and DNA, it prevents the religation of DNA strands, leading to double-strand breaks, cell cycle arrest in the S and G2 phases, and ultimately programmed cell death. It is primarily utilized as a precursor in the synthesis of more soluble and less toxic podophyllotoxin analogs for the treatment of various malignancies, though it is also listed within XinKe's pipeline for cancer applications.
