4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DISU), also commonly known as DIDS, is a small molecule research compound that acts as a potent inhibitor of the Hepatocyte Growth Factor Receptor (HGFR, also known as c-Met). In oncological research, particularly involving HT29 colorectal cancer cells, DISU is utilized to investigate the complex interplay between HGFR signaling, TLR9-mediated pathways, and autophagy. It has been shown to inhibit the HGFR-mTOR-ULK1 molecular cascade, thereby reducing cell proliferation and modulating autophagic flux. Beyond its application in oncology, DISU is a well-characterized, irreversible inhibitor of anion exchange proteins (such as the SLC4 family) and various chloride channels. It exerts its inhibitory effect by covalently reacting with amino groups, specifically lysine residues, on its target proteins.
