Drug intelligence / Profile preview
4-amino-1,8-naphthalimide 10-8-10 dimer (also known as 10-8-10 dimer) is a photoactivatable small molecule photosensitizer developed by Alucent Biomedical (originally spun out of Avera Research Institute) for use in Natural Vascular Scaffolding (NVS) therapy. Delivered locally to the blood vessel wall via a drug-coated balloon catheter during angioplasty or arteriovenous fistula (AVF) creation, the molecule is activated by 450 nm blue light delivered via an intravascular light fiber. Upon photoactivation, the dimer undergoes excitation to a triplet state, initiating a photochemical reaction that forms covalent crosslinks between oxidizable amino acid residues (such as tyrosine) in the native extracellular matrix (ECM) proteins, specifically collagen and elastin. This photochemical crosslinking reinforces the vessel wall, creating a natural biological scaffold that maintains the dilated lumen diameter, prevents acute elastic recoil, and promotes outward vascular remodeling without the need for a permanent metallic stent or foreign implant.
