Overview

This compound is a synthetic small molecule inhibitor of the bifunctional protein GlmU from Haemophilus influenzae. GlmU is an essential enzyme in bacterial aminosugar metabolism, catalyzing the last two sequential reactions in the de novo biosynthetic pathway for UDP-N-acetylglucosamine (UDP-GlcNAc). The inhibitor was identified through high-throughput screening and exhibits micromolar potency against hiGlmU. It binds to an allosteric site adjacent to the substrate-binding region, preventing structural rearrangements required for enzymatic activity. This mechanism provides a basis for structure-guided design of new classes of mechanism-based inhibitors targeting bacterial cell wall biosynthesis[5][10].
