Overview

5'-O-(L-Prolylsulfamoyl)adenosine is a synthetic small molecule and a purine nucleoside analog. This compound acts as a stable analog of the aminoacyl adenylate intermediate formed during tRNA aminoacylation by aminoacyl-tRNA synthetases. Specifically, it mimics the prolyl-AMP intermediate and serves as a potent inhibitor of prolyl-tRNA synthetase (ProRS), an enzyme essential for protein synthesis in cells[1][6][9]. Its primary use has been in biochemical and structural studies to investigate enzyme mechanisms rather than as an approved therapeutic agent.
