5'tRF-GlyGCC mimic is a synthetic RNA oligonucleotide designed to replicate the functional activity of the endogenous 5' transfer RNA-derived fragment (tRF) generated from tRNA-Gly-GCC. In breast cancer, this specific tRF is overexpressed and functions as an oncogenic driver. The mimic acts by binding to lactate dehydrogenase A (LDHA), particularly when LDHA is phosphorylated at tyrosine 10 by fibroblast growth factor receptor 1 (FGFR1). This binding enhances the enzymatic activity of LDHA, thereby promoting aerobic glycolysis and metabolic reprogramming that supports tumor cell malignancy. Additionally, the 5'tRF-GlyGCC/LDHA axis induces the secretion of the chemokine CCL7, which facilitates macrophage infiltration and polarization toward a pro-tumorigenic M2 phenotype. While the mimic is primarily used in research to characterize these oncogenic pathways, the axis it targets is being investigated for therapeutic intervention in breast cancer.
