Overview

Alpheus Medical is developing an investigational sonodynamic therapy (SDT) that combines oral 5-aminolevulinic acid (5-ALA) with a proprietary Low-Intensity Diffuse Ultrasound (LIDU) device for the treatment of glioblastoma. The therapy works through a two-step mechanism: 5-ALA is metabolized to protoporphyrin IX (PpIX), which selectively accumulates in glioblastoma tumor cells. When activated by diffuse ultrasound delivered across the entire brain hemisphere by the LIDU device, the PpIX generates reactive oxygen species (ROS) that trigger tumor cell death while sparing healthy brain tissue. This combination drug-device therapy is designed to be non-invasive and repeatable in an outpatient setting. It is currently being evaluated in Phase 2 clinical trials for newly diagnosed glioblastoma and has completed Phase 1 trials for recurrent glioblastoma.
