Overview

5-bromo-N,N-dimethyltryptamine (5-Br-DMT) is a naturally occurring brominated indole alkaloid found in marine sponges, such as *Smenospongia aurea*. It is a structural analog of N,N-dimethyltryptamine (DMT) that acts as a partial agonist at the serotonin 5-HT2A receptor and exhibits high affinity for several other serotonin receptor subtypes, including 5-HT1A, 5-HT2B, 5-HT2C, 5-HT6, and 5-HT7, as well as the serotonin transporter (SERT). Preclinical research characterizes 5-Br-DMT as a non-hallucinogenic psychoplastogen, as it promotes structural neuroplasticity (dendritogenesis) and upregulates plasticity-related immediate early genes (IEGs) without inducing the head twitch response in rodents. It has demonstrated rapid antidepressant-like effects in animal models of stress-induced depression, making it a candidate for the development of next-generation antidepressants that lack the intense psychedelic effects associated with traditional tryptamines.
