Overview

5-chloro-N,N-dimethyltryptamine (5-Cl-DMT) is a halogenated derivative of N,N-dimethyltryptamine (DMT) and a member of the tryptamine family of psychedelic compounds. It acts as an agonist at several serotonin receptors, most notably 5-HT1A, 5-HT2A, and 5-HT2C, and also inhibits the serotonin transporter (SERT). In preclinical studies, 5-Cl-DMT has been shown to induce the head-twitch response (HTR) in mice, which is a behavioral marker for hallucinogenic activity, although it exhibits lower potency and efficacy in this regard compared to 5-fluoro-DMT. The compound is being investigated for its psychoplastogenic properties, as it upregulates immediate early genes (IEGs) such as Arc and Egr-2 in the prefrontal cortex and hippocampus, indicating a potential for treating mood disorders by promoting structural neuroplasticity. It is currently utilized primarily as a research chemical and analytical reference standard.
