This is a combination therapy consisting of 5-fluorouracil (5-FU), a cytotoxic antimetabolite chemotherapy agent, and bevacizumab, a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A). 5-fluorouracil inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death in rapidly dividing tumor cells. Bevacizumab binds to VEGF-A, preventing it from activating its receptors on endothelial cells and thereby inhibiting angiogenesis—the formation of new blood vessels that tumors need for growth and metastasis. The combination is primarily used in the treatment of metastatic colorectal cancer, where it has been shown to improve response rates and survival when added to standard chemotherapy regimens[2][3][9].
