Overview

5-fluorouracil + carboplatin is a combination chemotherapy regimen consisting of two cytotoxic agents, 5-fluorouracil (also known as 5FU or fluorouracil) and carboplatin. This combination is primarily used to treat advanced cancers of the head and neck, as well as esophageal squamous cell carcinoma in patients who are ineligible for cisplatin-based therapy. Both drugs act by interfering with DNA synthesis and function, leading to inhibition of cancer cell proliferation. - **Mechanism of Action:** - 5-fluorouracil is an antimetabolite that inhibits thymidylate synthase, thereby blocking DNA synthesis. - Carboplatin is a platinum-based compound that forms DNA crosslinks, resulting in apoptosis due to impaired DNA replication and transcription. This regimen may be considered when cisplatin cannot be used due to toxicity concerns. The combination has demonstrated comparable efficacy to cisplatin/fluorouracil regimens but with a different side effect profile[1][3][4].
