This is a combination regimen consisting of three drugs: - **5-fluorouracil (5-FU):** A pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. It is commonly used as chemotherapy for various cancers. - **Cetuximab:** A chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling pathways involved in tumor cell proliferation and survival. - **Oxaliplatin:** A platinum-based chemotherapeutic agent that forms DNA crosslinks, inhibiting DNA replication and transcription, resulting in apoptosis. This combination has been studied primarily as a first-line treatment for metastatic colorectal cancer, particularly in patients with EGFR-expressing tumors. The regimen may be administered with or without folinic acid (leucovorin) depending on protocol. Clinical trials have shown mixed results regarding its efficacy; some studies suggest benefit in KRAS wild-type tumors while others do not show significant improvement over standard regimens[1][2][3].
