This combination leverages multiple cytotoxic mechanisms plus targeted therapy against HER2-positive cancers. It has been studied in various cancers including breast cancer and HER2-positive esophagogastric adenocarcinoma. Trastuzumab addition improves survival outcomes in HER2-positive tumors when combined with chemotherapy[6][8][10]. The regimen includes drugs from different classes such as alkylating agents, antimetabolites, taxanes, anthracyclines, and monoclonal antibodies.
