5-fluorouracil + CoFactor

5-fluorouracil + CoFactor is a combination therapy consisting of the antimetabolite 5-fluorouracil (5-FU) and the folate modulator CoFactor (ANX-510). CoFactor is the active form of folate, 5,10-methylenetetrahydrofolate, which does not require the metabolic conversion steps needed by leucovorin. It works by stabilizing the ternary complex between the 5-FU metabolite fluorodeoxyuridylate (FdUMP) and the enzyme thymidylate synthase. This stabilization leads to more effective and prolonged inhibition of thymidylate synthase, thereby depleting thymidine pools and inhibiting DNA synthesis and repair. Developed by Mast Therapeutics (formerly Adventrx Pharmaceuticals), this combination was primarily investigated for the treatment of advanced colorectal cancer and metastatic breast cancer, particularly in patients who had failed prior standard-of-care therapies such as anthracyclines and taxanes.
