This is a combination therapy consisting of 5-fluorouracil (5-FU), a widely used antimetabolite chemotherapy agent, and Corynebacterium parvum, a bacterial immunostimulant. - **5-Fluorouracil** acts as an antimetabolite by inhibiting thymidylate synthase, thereby interfering with DNA synthesis and cell division in rapidly proliferating cancer cells[6][7]. - **Corynebacterium parvum** is used as an immunopotentiator; it stimulates the immune system non-specifically to enhance anti-tumor responses. The rationale for combining these agents is to leverage both direct cytotoxic effects on tumor cells (from 5-FU) and immune-mediated anti-tumor activity (from C. parvum)[1][7]. Clinical studies have evaluated this combination in advanced cancers such as colorectal carcinoma and breast cancer. However, results have shown limited or no additional therapeutic benefit over chemotherapy alone in some settings[7], while other studies suggest possible enhancement of tumor inhibition depending on the chemotherapeutic partner[1].
