5-fluorouracil + dipyridamole is a chemotherapy combination where dipyridamole acts as a modulator to enhance the cytotoxicity of 5-fluorouracil. Dipyridamole, an inhibitor of nucleoside transport, increases the activity of 5-fluorouracil in a dose-dependent manner. The combination has been studied in Phase I and II clinical trials for various cancer types, including advanced refractory malignancies, breast cancer, and transitional cell carcinoma. In clinical studies, dipyridamole has been administered both orally and intravenously in combination with 5-FU, often with the addition of folinic acid (leucovorin) to further enhance efficacy.
