Overview

This is a combination chemotherapy regimen used primarily as first-line treatment for metastatic breast cancer. The regimen consists of three drugs administered intravenously on an outpatient basis every three weeks. Vinorelbine is a vinca alkaloid that disrupts microtubule formation, inhibiting cell division. 5-Fluorouracil (5-FU) is an antimetabolite that interferes with DNA synthesis by inhibiting thymidylate synthase, especially when combined with folinic acid (leucovorin), which enhances the cytotoxic effect of 5-FU by stabilizing the binding of its active metabolite to thymidylate synthase[1][2]. The combination has demonstrated efficacy comparable to other first-line regimens for advanced breast cancer, with manageable toxicity profiles[1][2].
