5-fluorouracil + leucovorin + panitumumab

A combination therapy consisting of 5-fluorouracil (a pyrimidine analog antimetabolite), leucovorin (a reduced folate that enhances 5-FU activity), and panitumumab (a fully human monoclonal antibody targeting EGFR). This regimen is primarily used for the treatment of metastatic colorectal cancer, particularly in patients with RAS/BRAF wild-type tumors. The combination works through complementary mechanisms: 5-FU inhibits thymidylate synthase and disrupts DNA synthesis, leucovorin enhances 5-FU's binding to its target enzyme, and panitumumab blocks EGFR signaling to inhibit tumor cell proliferation and survival.
