This is a combination chemotherapy regimen consisting of three cytotoxic agents: - 5-fluorouracil (an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis) - Lomustine (also known as CCNU, an alkylating nitrosourea that crosslinks DNA and RNA) - Vincristine (a vinca alkaloid that inhibits microtubule formation in mitosis) The combination has been studied primarily as adjuvant or palliative therapy for colorectal cancer, particularly in the postoperative setting for Dukes' C colorectal cancer. The regimen aims to target tumor cells through multiple mechanisms—disrupting DNA synthesis and function as well as inhibiting cell division. However, clinical trials have shown no significant improvement in survival compared to no adjuvant therapy in this context[1][3][4]. Side effects are common and can lead to discontinuation of treatment.
