This is a combination therapy consisting of three agents: - **5-fluorouracil** (5-FU): A pyrimidine analog antimetabolite chemotherapy that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. It is widely used in the treatment of colorectal and other gastrointestinal cancers. - **TroVax**: A cancer vaccine developed by Oxford BioMedica, based on a modified vaccinia virus Ankara (MVA) vector encoding the tumor-associated antigen 5T4. TroVax aims to stimulate an immune response against tumors expressing 5T4, which is present in many solid tumors including colorectal cancer[5][7]. - **Oxaliplatin**: A platinum-based alkylating agent that forms DNA crosslinks, inhibiting DNA replication and transcription, resulting in apoptosis of rapidly dividing cells. It is commonly used with 5-FU for advanced colorectal cancer[6][8]. The combination leverages both cytotoxic chemotherapy (oxaliplatin and 5-FU) and immunotherapy (TroVax), aiming for synergistic anti-tumor effects by directly killing tumor cells while also stimulating an immune response against them.
