Overview

A combination of two dietary supplements—5-hydroxytryptophan (5-HTP), a serotonin precursor, and creatine monohydrate, a compound involved in cellular energy metabolism. This combination is being investigated as an adjunctive treatment for major depressive disorder (MDD), particularly in individuals who have not responded adequately to selective serotonin reuptake inhibitors (SSRIs) or serotonin-norepinephrine reuptake inhibitors (SNRIs). The rationale is that 5-HTP may increase central serotonin synthesis while creatine may enhance brain bioenergetics. Early clinical studies suggest this combination can reduce depressive symptoms when added to standard antidepressant therapy, with good tolerability over short-term use[1][2][3][4][8].
