Overview

INC2 (5-iodoindirubin-3'-monoxime) is a synthetic derivative of indirubin, a bis-indole alkaloid found in indigo-producing plants and the traditional Chinese medicine Qing Dai. Evaluated primarily in the context of dermatology by the CRO Derphartox, INC2 acts as a potent inhibitor of cyclin-dependent kinases (CDKs), including CDK1, CDK2, and CDK5, as well as glycogen synthase kinase-3 (GSK-3α/β). In preclinical models of psoriasis, such as the Derphartox skin transplant model, INC2 demonstrates anti-proliferative effects on human keratinocytes and modulates inflammatory signaling pathways, including the inhibition of STAT3 activation and interaction with the aryl hydrocarbon receptor (AhR). It is frequently utilized as a positive control or reference compound in studies assessing novel therapeutic candidates for psoriasis and other hyperproliferative skin disorders.
