5-MAPB + 6-APB is a preclinical-stage small molecule drug combination being developed by Clearmind Medicine for use in drug-assisted psychotherapy. The combination consists of two benzofuran-class entactogens: 5-MAPB (1-(benzofuran-5-yl)-N-methylpropan-2-amine) and 6-APB (6-(2-aminopropyl)benzofuran). These compounds are structurally and pharmacologically related to MDMA and act as triple monoamine reuptake inhibitors and releasing agents, targeting the serotonin, dopamine, and norepinephrine systems. Clearmind is investigating this combination as a potential 'fail-safe' or alternative to MDMA-assisted psychotherapy, particularly for patients who have developed tolerance to MDMA or for whom MDMA therapy is ineffective or inappropriate. Primary targeted indications include post-traumatic stress disorder (PTSD), generalized anxiety disorder (GAD), and relational or relationship-based psychological distress.
