Overview

6-(2,3-difluoro-5-((methylamino)methyl)phenyl)-4-methylpyridin-2-amine is a potent and selective small molecule inhibitor of neuronal nitric oxide synthase (nNOS). It belongs to the class of truncated pyridinylbenzylamines and was developed as part of efforts to create highly membrane-permeable inhibitors targeting nNOS. The compound demonstrates excellent potency with low nanomolar K_i values against human nNOS and shows selectivity over other nitric oxide synthase isoforms such as endothelial NOS (eNOS). Its mechanism involves direct binding to the active site of nNOS, thereby inhibiting its enzymatic activity. This makes it a promising lead for therapeutic development in neurological disorders where aberrant NO signaling is implicated, as well as for melanoma[1][2][3].
