Overview

6-(adenosine tetraphosphate-methyl)-7,8-dihydropterin is an experimental small molecule bisubstrate analogue designed to inhibit the enzyme 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase (HPPK). HPPK catalyzes a key step in folate biosynthesis by transferring pyrophosphate from ATP to 6-hydroxymethyl-7,8-dihydropterin. This enzyme is essential for microorganisms but absent in humans and animals. The compound consists of a pterin moiety linked via four phosphate groups to an adenosine moiety. It acts as a potent competitive inhibitor of HPPK by occupying both the pterin and ATP binding sites on the enzyme. Its high affinity for HPPK makes it valuable as a tool compound for biochemical studies and as a lead structure for antimicrobial drug development targeting folate biosynthesis in bacteria[1][3][2].
