(64Cu)-NOTA-PSMAi-PEG-Cy5.5-C' dot tracer is an investigational dual-modality imaging agent consisting of ultrasmall silica nanoparticles (C' dots) functionalized for both positron emission tomography (PET) and intraoperative fluorescence imaging. Developed by Memorial Sloan Kettering Cancer Center in collaboration with Cornell University, the tracer incorporates a prostate-specific membrane antigen (PSMA) inhibitor for targeting, a PEG linker for stability, the near-infrared fluorescent dye Cy5.5, and the chelator NOTA for labeling with the radioisotope copper-64 (64Cu). It is designed to improve the localization of prostate cancer deposits during surgery by providing pre-operative PET/MRI scans and real-time intra-operative optical guidance. The ultrasmall size of the silica platform facilitates favorable biodistribution and renal clearance, while the dual-labeling allows for a theranostic-like approach to surgical management.
