Drug intelligence / Profile preview
64Cu-porphysomes are inherently multimodal, all-organic nanoparticles composed of self-assembled porphyrin-lipid conjugates. Developed by the University Health Network (UHN), these nanovesicles (porphysomes) utilize the intrinsic metal-chelating properties of the porphyrin bilayer to stably incorporate Copper-64 (64Cu), enabling high-sensitivity positron emission tomography (PET) imaging. Beyond radiotracing, the nanoparticles exhibit unique biophotonic properties, including structure-dependent fluorescence self-quenching and photoacoustic signaling, which allow for multimodal tumor delineation. They primarily accumulate in cancerous tissues through the enhanced permeability and retention (EPR) effect. Clinical development is focused on the imaging and staging of advanced metastatic gynecological cancers, while preclinical studies have demonstrated efficacy in prostate cancer models.
